The fastest way to earn points to enter the cyclops roo is to reanimate armour on the ground floor. Try to aim for around 1k tokens, that should be enough to get the dragon defender depending on RNG

Once in the cyclops room all defender drops are uncommen drops, make sure you show you're defender to Kamfreena everytime you get a new one. once you have a rune defender you have a chance for a cyclossus to spawn but only if you are wearing an rune/dragon defender will they be able to spawn and receive damage.
