Once in the cyclops room all defender drops are uncommen drops, make sure you show you're defender to Kamfreena everytime you get a new one. once you have a rune defender you have a chance for a cyclossus to spawn but only if you are wearing an rune/dragon defender will they be able to spawn and receive damage.
